The sample comprising the target nucleic acid is lysed e.g. by using a lysis buffer, mechanical lysis, physical lysis or biological lysis by using appropriate enzymes if necessary. Preferably, the sample is disrupted in the presence of a lysis buffer. Afterwards, a complex of target nucleic acid (e.g. DNA,RNA) and a silica based matrix is formed upon binding of the target nucleic acid to the nucleic acid binding matrix. After wash columns with wash buffer1 and wash buffer2,then elution the DNA,RNA with pure water or TE.

The Spin column bind DNA and RNA with Similar efficiency,also short fragment nucleic acid(-80bp) and long fragment eg.genomic DNA .

* Storage

Spin columns should be stored at 2–8°C upon arrival and are stable under
these conditions for at least one year after delivery. However, short-term storage of up
to 8 weeks at room temperature (15–25°C) does not affect performance.
